1. Design scale and principles

The scale of plant tissue culture laboratory design depends on the purpose of the work, such as factory production or scientific research.

The design principles are as follows:

1) Ensure aseptic operation: Plant tissue culture is carried out under strict aseptic conditions. To achieve sterile conditions, certain environments, equipment, equipment and utensils are required.

2) Work convenience: Follow the tissue culture procedures to avoid back-ordering certain links and causing confusion in future work.

3) Culture conditions: artificial control of temperature, light, humidity, etc.

2. Laboratory composition

The standard tissue culture room includes drug area, washing area, preparation area, sterilization area, storage area, sterile operation area and culture area, etc., which can be combined and established according to your own actual conditions.

3. Laboratory planning and design

1) Preparation room

Main tasks: storage, weighing, dissolution, preparation, and culture medium packaging of drugs. Washing, drying, and preservation of utensils, sterilization of culture media, etc.

Main equipment: medicine cabinet, dust-proof cabinet (for placing culture containers), refrigerator, electronic balance, distilled water device, acidometer, high-pressure sterilizer, drying sterilizer (such as oven), commonly used glass instruments for culture medium preparation, operation Taiwan etc.

2) Inoculation room (sterile operating room)

Main tasks: Mainly used for disinfection of plant materials, inoculation, transfer of cultures, subgeneration of test tube seedlings, preparation of protoplasts and all technical procedures requiring aseptic operations.

Main equipment: UV light source, ultra-clean workbench, sterilizer, alcohol lamp, vaccination equipment (vaccination tweezers, scissors, scalpel, vaccination needle), etc.

The inoculation room should be small rather than large, generally 7 to 8 square meters. The floor, ceiling and walls are required to be as airtight and smooth as possible, and easy to clean and disinfect. Configure the pull door to reduce air disturbance when opening and closing the door.

The vaccination room must be dry, quiet, clean and bright. Hoist 1 to 2 ultraviolet sterilization lamps at appropriate locations for irradiation and sterilization. Install a small air conditioner to control the room temperature, so that the doors and windows can be closed tightly to reduce convection with outside air.

The inoculation room should be equipped with a buffer room, with an area of 1 square meter. Change clothes and shoes before entering the sterile operating room to reduce the number of germs brought into the inoculation room when entering and exiting. An ultraviolet sterilization lamp is also installed in the buffer room for irradiation and sterilization.

3) Training room

The culture room is a place where inoculated materials are cultured and grown.
Determine the area according to the size, number of culture racks, and other ancillary equipment required. Its design is based on the principles of making full use of space and saving energy. The height should be slightly higher than the culture rack, and the surrounding walls must have thermal insulation and fireproof properties.

The important factor in the culture room is the temperature, which is generally maintained at around 20-27°C. It must have a heat-generating device and install a window or vertical air conditioner.

The culture rack is generally set up in 5 layers. The lower layer is about 10cm high from the ground. The other layers are spaced about 30cm apart. The height of the culture rack is about 1.7m. The length is designed according to the length of the fluorescent lamp. If a 40W fluorescent lamp is used, the length is 1.3m, the 30W fluorescent lamp is 1m long, and the width is generally 60cm.

Since different species such as tropical plants and boreal plants require different temperatures, different species have different culture rooms. The indoor humidity also needs to be constant. The relative humidity should be maintained at 70 to 80%. A humidifier can be installed. A timer switch clock can be installed to control the lighting time. Generally, 10-16 hours of lighting are required every day, and some require continuous lighting.

Most modern tissue culture laboratories are designed to use natural sunlight as the main energy source. This not only saves energy, but also allows tissue culture seedlings to grow well under sunlight and be easily acclimated and survive. Lights can be used as a supplement on rainy days.

Main equipment: culture rack (temperature control, light control and humidity control), shaker, incubator, UV light source, vertical air conditioner, etc.

4) Cytology laboratory

This chamber is used for observation and analysis of cultures and counting of cultures.

Main equipment: binocular solid microscope, microscope, inverted microscope, etc. Small instruments and equipment include dispensers, hemocytometers, pipette guns, filter sterilizers, electric furnaces and other heating appliances, magnetic stirrers, low-speed desktop centrifuges, etc.
